Molecular characterization and genetic diversity analysis of Turkish bottle gourd (<i>Lagenaria siceraria</i> (Molina) Standl.) genotypes using SRAP, ISSR and SSR markers

Bottle gourd (Lagenaria siceraria (Molina) Standl.) is an important cucurbit species used as a vegetable crop and as a rootstock in grafted watermelon production. This study evaluated the genetic diversity and relationships among 40 bottle gourd genotypes collected from southern T & uuml;rkiye using sequence-related amplified polymorphism (SRAP), inter-simple sequence repeat (ISSR) and simple sequence repeat (SSR) markers. The three marker systems generated 468 scorable DNA fragments, of which 344 were polymorphic, corresponding to an overall polymorphism rate of 73.5%. SRAP markers produced 201 fragments with 74.1% polymorphism, ISSR markers generated 202 fragments with 72.8% polymorphism and SSR markers produced 65 fragments with 73.8% polymorphism. Dice similarity coefficients ranged from 0.370 to 0.988, indicating substantial variation in molecular relatedness among the genotypes. Unweighted pair group method with arithmetic mean (UPGMA) clustering separated the genotypes into two unequal groups comprising 34 and six genotypes, while principal coordinate analysis (PCoA) provided a complementary ordination of the genetic relationships and showed partial concordance with the dendrogram. Bayesian population structure analysis supported K = 2 and identified five genotypes showing mixed membership coefficients below 0.80. The cophenetic correlation coefficient (r = 0.836) indicated a good representation of the original distance matrix by the dendrogram. Genotypes SK1 and SK5 were the most divergent, whereas SK25 and SK35 showed the highest similarity. The results demonstrate that Turkish bottle gourd germplasm contains useful molecular variation and that the combined use of SRAP, ISSR and SSR markers is an effective approach for germplasm characterization, conservation and parental selection in future breeding programs.
